I am suffering from low platelet count since three years. My recent count on automatic analysis is 70000 and manual platelet count is 100000. Which result is more significant and rest blood report is within normal parameters. What could be the reason?

Low platelet count since three years indicates blood dyscrasia and need detailed check up. You do a PBS test and iform. May need to do bone marrow test to help diagnose. If the manual count is normal it is more reliable and automated results are not always correct.
