I've observed a practice in my relatives home that they don't throw the the tea leaves after first use and they reuse it for one more time same day. Is it fine? Or we should not use the used leaves at all? Or we can use them within 3-4 hours after first use?

Please It is not good Actually we should not overboil tea leaves while making tea Reused tea leaves are toxic for health.

Dear user. You can see this practice in many tea vendor shops too. There is nothing wrong in using tea leaves for a second or third time. But the quality will be less. Take care.
