If the doppler study is normal and not showing any thrombosis (clot), it may be an age-related pain.
There may not be any relation between the covid vaccine and the leg pain because having pain after 40 days of the vaccination is unlikely due to adverse effect of the vaccination.
Consult an orthopedician for better assessment and she may have to do physiotherapy.

The doctors must have suspected pulmonary embolism. Running test like D Dimer has got no role in diagnosing this in non ambulatory patients. Please go for CECT thorax to have an idea of the condition. Is your brother on heparin or warfarin therapy?

Protein s deficiency is a hyper coagulable state which will lead to miscarriages if not treated. Low molecular weight heparin like dalteparin (fragmin) is a treatment for protein s deficiency that you will need to take.
