Diphosphate nucleoside inhibits ribonucleotide reductase which generates deoxynucleoside triphosphates which are essential for DNA synthesis. Thus, the gemcitabine triphosphate competes with deoxynucleoside triphosphates for incorporation into DNA and interferes with DNA synthesis, results in cell death.
