KLB Smear test is used to measure the level of monochromatic granules in the throat swab, This test is performed to detect Corynebacterium infection, Nasopharyyngeal lesions and Skin lesions. Corynebacterium causes diphtheria which is a serious bacterial infection usually affecting the mucous membranes of your nose and throat. Diphtheria typically causes a sore throat, fever, swollen glands and weakness. But the hallmark sign is a sheet of thick, gray material covering the back of your throat, which can block your airway, causing you to struggle for breath.

This test is used to determine the level of monochromatic granules present in throat swab. With the help of this test one can deduct the presence of Corynebacterium Which causes diphtheria.

The sample is collected from the Pharyngeal wall by a sterile cotton fitted to a 6-7 inches long swab stick. The cotton is rubbed along the pharyngeal mucosa. Precaution is to be taken to avoid the sample being contaminated. The sample is then transferred to a small sterile vial containing saline solution. Culture medium used is Nutrient Agar, Blood Agar, Mackconkeys Agar and Loeffler’s serum.
