My right thais muscles above the knee has grown fat. Sitting and standing has become very painful it gives a lot of pain on right thai till knees.

Hello lybrate-user, pain while sitting and standing mostly due to quads weekness which may be due to osteoarthritic changes or some other cause need to in details about age, pain duration, trauma ,knee movement problem, sound while movement. If only pain while sitting and standing I would suggest start quadriceps strengthening exercises.

You need to do proper stretching and strengthening exercises for hamstrings, quadriceps and piriformis. Also do core strengthening for hip, back and abdominal muscles. This will reduce fat, pain and help tone thigh muscles.
